UT-A1 Interacting Proteins in Rat Inner Medullary Collecting Duct

This database reports proteins identified by mass spectrometry (LC-MS/MS) that bound to urea channel protein UT-A1 in rat inner medullary collecting duct. IMCD suspenions were treated with vehicle or vasopressin analog (dDAVP, 1 nM) for 15 minutes, followed by in-cell crosslinking using BSOCOES. After incubations, IMCD were harvested by brief centrifugation. The samples were homogenized and solubilized with RIPA. The detergent solubilized fractions were subjected to immunoprecipitation (IP) using UT-A1 antibody-coated Dynabeads or preimmune IgG-coated beads as control. The IP products were eluted in Laemmli buffer and loaded for SDS-Page. Gel pieces corresponding to UT-A1 high molecular weigh complex band above 250 kDa were cut and prepared for mass spectrometry (LC-MS/MS) analysis according to standard in-gel digestion procedures. Proteins that found in all three experiments and were statistically significant in either control or dDAVP-treated group were included in the interactome list.

Definitions: R, ratio (peptide intensity in UT-A1 antibody specific immunoprecipitation sample)/(peptide intensity in preimmune-IgG immunoprecipitation sample) for each peptide found. INF, infinite value. p-value, P value of student t-test vs log2R=0. -log (P) greater than 1.30 was considered statistically significant. S, significant, P value was not available. NS, not significant. -, not available.
