11226305. Methods: Mouse mpkCCDcl4 cells were used, a collecting duct-derived transformed cell line.Cells were treated with 10 nM vasopressin (Vaso4h) for 4 h or with vehicle. The mRNAs of the control and and Vaso4h conditions were prepared in two independent sets of confluent cells grown on filters having identical electrical properties. Poly(A) mRNAs were selected and SAGE libraries were prepared from 5 μg of the corresponding poly(A) mRNAs. 10,000 sequencing reactions were run. Assessment of significant differences between two SAGE libraries was made by Monte Carlo simulation analysis and validated by a Z-test. A P value of 0.05 or less was considered significant. Transcripts significantly increased by vasopressin are labeled as VITs. Transcripts significantly decreased by vasopressin are labeled VRTs. The identifications were re-annotated in 2014 based on current RefSeq data. |
