PMID: 17913837. Methods: AQP1 knockout mice were infused with the V2-selective vasopressin analog dDAVP via osmotic minipumps at 0.5 ng/h for 7 days. Controls received vehicle. RNA samples were isolated from individual mouse inner medullas. After reverse transcription and amplification the cDNAs obtained were fluorescently labeled using 546 Alexa Fluor and 647 Alexa Fluor for dDAVP/control pairs. Relative transcript levels were read out using cDNA arrays constructed using the National Institute on Aging mouse 15K clone set http://lgsun.grc.nia.nih.gov/cDNA/15k.html. (AQP1 knockout mice were used to prevent large changes in medullary osmolality which could have been a confounding factor. AQP1 is necessary for countercurrent multiplication.) Data are mean values from three pairs of samples from different mice. |
