This database contains protein mass spectrometry data from the NHLBI Epithelial Systems Biology Laboratory (ESBL). Data are obtained from the late distal convoluted tubule (DCT2), the connecting tubule (CNT), and the initial cortical collecting duct (iCCD) from aldosterone treated mice.
Experimental description: Transgenic mice expressing eGFP in the distal tubule principal cells were treated with 2 mg/kg aldosterone 24 hours prior to sacrifice. After enzymatic digestion, DCT2, CNT, and iCCD cells were isolated based on their endogenous fluorescence properties using Fluorescent Activated Cells Sorting (FACS). Isolated cells were sonicated on ice in 8M urea. All mass spectrometry experiments were conducted using isobaric Tags for Relative and Absolute Quantitation (iTRAQ) which enables quantitation of a large number of peptides from several samples. The samples were run on a mass spectrometer (LTQ Orbitrap Velos). Normalized iTRAQ values for vehicle and aldosterone groups are reported for each protein.
